How we can load bootloader program into it ?

you can program the micro controller with boot loader by using any burner (programmer)

Yes ISP programmer, bootloader is only needed if you want to support system update on field. If you are going to do firmawre upgrade on desk then ISP is enough no need for bootloader.

I want to load HEX file(program) into microcontroller using LabVIEW ..How i can achieve this??

Labview should be supporting AVR isp programming. I dont think bootloader is needed in this case.

You will need to use ISP programmer. I think better way will be to generate hex file from labview and then program it with any AVR programmer like AVRDude or USBASP, just search for "AVR Programmer" on google you will find many of them.
